• W2980821377 abstract "Neurofilament-light (NfL) and total tau (T-tau) in cerebrospinal fluid (CSF) have both been identified as neurodegenerative markers in Alzheimer's disease (AD), but also in other neurodegenerative disorders like cerebrovascular disease (CVD). Aim of this study was to investigate whether the associations of NFL and T-tau with cerebrovascular magnetic resonance imaging (MRI) markers is dependent on the presence of amyloid pathology. We selected 419 individuals with normal cognition (NC, n=93) or mild cognitive impairment (MCI, n=326) from the multi-center EMIF-AD Multimodal Biomarker Discovery study. CSF NfL and amyloid-β42/40 (Aβ42/40) ratio were analyzed centrally. Amyloid status (A -/+) was defined by the Aβ42/40 ratio with a cut-off of <0.063. CSF T-tau was analyzed locally and standardized by assay type. Hippocampal volume (HCV), white matter hyperintensities (WMH), microbleeds and lacunes on MRI were analyzed centrally using automated volumetric analysis and visual rating. Spearman correlations were used to determine associations between the markers. Individuals were on average 68.1 (SD 8.0) years old and 52% were female. Table 1 shows the correlations between the CSF and MRI markers. Among the individuals with NC, we found that NfL was associated with lower HCV and a higher number of microbleeds in the A+ group. T-tau was only associated with lower HCV in the A- group. In individuals with MCI, we found that NfL was associated with lower HCV, more severe WMH and a higher number of lacunes, regardless of amyloid status. NfL was also associated with more microbleeds but only in the A+ group. T-tau was associated with lower HCV and more severe WMH, but only in the A- group." @default.
