- W2981292163 abstract "Cerebral deposition of amyloid can occur decades before the expression of Alzheimer's disease. Recent research suggests that changes in amyloid deposition can be detected as a function of estimated years to onset (EYO) in persons with family history (FH) of sporadic Alzheimer's disease (AD). Carrying the ApolipoproteinE ε4 allele (APOE-ε4) accelerates amyloid deposition. The objective of the study was to evaluate the independent and interactive effects of APOE-ε4 status and EYO on AD biomarkers in cognitively unimpaired (CU) middle-aged adults. We included 204 CU middle-aged adults with FHAD (age 49-73, 63% females, 40% APOE-ε4 carriers) from the ALFA study. EYO was calculated as the age of the participant at assessment minus the age of the parent at symptom. 18F-flutemetamol-PET SUVRs, CSF Aβ42 and phosphorylated tau (p-tau) were used as AD biomarkers. We performed ANCOVA models, including AD biomarkers as dependent variables and APOE-ε4 status (carrier/non carrier), sex, age, education and EYO as independent variables. We also ran sensitivity analyses to test whether paternal/maternal history, sex and low education (<12 yrs) modified the effects. Proximity to parental EYO was associated with increased 18F-flutemetamol-PET SUVRs (F=4.53, p=.035) but not CSF Aβ42 (p=.19) or p-tau (p=.65). When APOE-ε4 was included in the model, both APOE-ε4 (F=4.59 p=.03) and EYO (F=4.18 p=0.04) showed effects on 18F-flutemetamol-PET after controlling for age, sex and education (Fig1A). There was no EYO* APOE-ε4 interaction (p=0.5).
