- W2981302848 abstract "Neurogranin (Ng) is a 78 amino acid, post-synaptic protein expressed mostly in neuronal (Represa et al., 1990), but also in some non-neuronal cells (Glynne et al., 2000;Gnatenko et al., 2003). Ng has been suggested to bind intracellularly to calmodulin and phosphatidic acid via its IQ domain and plays an important role in long-term potentiation (Zhong et al., 2009). It is detected at increased concentration in CSF from patients with Alzheimer's disease, as compared to age-matched controls (Thorsell et al., 2010); it is present as full-length protein and C-terminal truncated peptides in CSF (Kvartsberg et al., 2015). In an attempt to identify the molecular forms of Ng in CSF, we performed ultrafiltration to concentrate pooled CSF followed by size exclusion chromatography, SDS-PAGE and immunoblotting. To determine, the ratio of C-terminal Ng fragments to total-Ng we immunodepleted full-length and N-terminal Ng using an antibody specific against N-terminal Ng and followed the depletion of the Ng signal by an in-house sandwich ELISA using antibodies specific against C-terminal Ng. Our results from pooled CSF show that Ng is present in higher molecular forms in addition to monomeric Ng and as C-terminal fragments. The results from CSF samples immunodepleted of full-length and N-terminal Ng suggest that C-terminal peptides are about 50% of total-Ng in CSF. Taken together, this study highlights the presence of several molecular forms of Ng in CSF that may either represent Ng monomers and oligomeric forms of it, or complexes of Ng with as yet unidentified binding partners. In addition to monomeric and higher molecular forms of Ng, C-terminal peptides are major fragments detected in CSF." @default.
