- W2981799109 abstract "Abstract Malaria is a mosquito-transmitted infectious disease caused by the protozoan parasite Plasmodium spp. that often leads to severe complications and death. The complex life cycle of parasites and the parasites’ ability to acquire immune escape strategies make it challenging to develop effective vaccines and antimalarial drugs. With over 200 million infection cases annually and the rising problem of multidrug resistance and the limited efficacy of vaccine candidates in clinical trials, we are still far from global malaria eradication. The design of vaccine for malaria is not restricted to protection against infection and transmission blocking, which is still a challenge for the current vaccine candidates. The control of disease severity and pathology is also an important aspect to reduce the burden of malaria. New approaches have to be considered to target the disease by modulating host immunity with wider perspectives of the choice of antigens, adjuvants, and immunization routes. Recent studies have revealed a bidirectional relationship between the gut microbiota and malaria infection: Malaria infection modulates mucosal immunity, and the gut microbiome also contributes to the disease susceptibility. It is therefore rational to suggest the potential of nasal or oral vaccination to elicit both local and systemic immunity for protection against infection and the disease pathology. This chapter will review the efforts of mucosal vaccination strategies against malaria in the context of host–Plasmodium interactions in the gastrointestinal system." @default.
