- W2996199135 abstract "Enzyme is a biomolecule that has the ability to catalyze a chemical reaction without reacting. One enzyme widely applied is Lipase Enzyme. Increased demand for lipase enzymes each year encourages research to obtain sources of lipase especially from plants. One potential source of lipase developed is coconut houstorium. This study aims to determine purified lipase enzyme by salting out method and to know lipase enzyme activity after purified by salting out method. The stage of the research was isolation of crude extract of lipase enzyme, fractionation with ammonium sulphate, protein content test, activity test and molecular weight determination through SDS-PAGE and Native-PAGE. The results showed that there was an increase in lipase enzyme activity after the fractionation process showed that the enzyme was pure. The results showed that lipase enzyme was found in all fractions with the highest activity found in the 0-30% fraction of 2.111 units/mL. The activity contained in each fraction of purification result is higher compared with crude extract of lipase before purified ie equal to 0,1222 U / mL. The highest specific activity was found in the 0-30% fraction of 3.059 units / mg and the specific activity of the lowest lipase enzyme was found in the 75-90% fraction of 0.446 units/mg. The SDS-PAGE results showed a thick band of 72.594 kDa molecular weight, while the Native-PAGE result showed a single band with a molecular weight of 90.99 kDa." @default.
