- W2999194193 abstract "ABSTRACT Introduction Oxaliplatin is an active integral component of adjuvant and palliative therapy of GIT cancers. Sensory neurotoxicity is dose limiting toxicity of Oxaliplatin; it is thought to be due to an effect on neural voltage –gated Na channels resulting from the liberation of one Oxaliplatin metabolites; particularly oxalate metabolite. The purpose of this study was to evaluate Neurotoxic adverse effect of Oxaliplatin weekly dose versus Bi/Tri weekly doses in Oxaliplatin based regimens. Methods A prospective study was conducted in a total of thirty –four patients who were admitted at Cairo Cure Oncology Center with different GIT tumors who received different doses of Oxaliplatin –based regimens, Arm A was composed of (11 patients) who received Oxaliplatin (OXA) at 40-45 mg/m2 (D1.8 & 15),while Arm B was composed of (23 patients) who received OXA –based regimens with dose ranges of 85mg/m2/2weeks and 120-135mg/m2 /3week; Neurotoxicity was assessed and graded according to NCI CTC V4 in both groups. The 2 Arms were comparable in terms of History of Diabetes Mellitus; 3 patient in Arm A(27%) and 8 patient in the Arm B (35%) Results The median age of our patients was (56) years. (3) Patients from Arm A suffered from Peripheral neuropathy Grade (I-II) (27%), while (10) patients from Arm B suffered from Peripheral neuropathy Grade ( II-III) (43%). Conclusion According to our data; Less Neurotoxicity is numerically observed in Weekly Oxaliplatin based regimens, However due to small sample size the difference was not statistically significant. Accordingly; large prospective randomized well controlled studies should be considered to further evaluate the efficacy and toxicity of Oxaliplatin schedules in OXA-Based regimens (Weekly versus Bi/Tri weekly doses)." @default.
