- W3009668085 abstract "Abstract When acetylcholine(ACh) is released, there is a compensatory increase in ACh synthesis, which has been suggested to be associated with an increase in high affinity choline uptake activity. However, the relationship between the metabolism of ACh and phospholipid is unclear. We found that choline was released from the synaptic vesicle fraction of rat brain through the following three pathways: (1)hydrolysis by alkaline phosphatase of phosphorylcholine, (2)enzymatic degradation of phosphatidylcholine by phospholipase A and glycerophosphorylcholine phosphodiesterase, and (3)by phospholipase D. A negative result was obtained on base exchange activity. In the synaptosomal fraction from rat brain incubated in a choline-free medium, the ACh synthesis was inhibited by 1-p-bromotetramisole, a selective and potent inhibitor for pathway (1), at 0.5mM by approximately 50 per cent, and by DFP for pathway (2) by approximately 10 percent Furthermore, in some experiments on insect ganglia perfused with choline-free saline solution, a releasable pool of transmitter was reduced in the presence of 0.1-0.5mM 3 chloro-4-stilbazole, an inhibitor for choline acetyltrans-ferase, whereas ImM HC-3 had no effect on it. These results suggest that there is some choline supply from endogenous phospholipids to ACh synthesis within cholinergic terminals." @default.
