• W3017629516 abstract "قارچ¬ها از جمله منابع طبیعی هستند که از کاربرد مختلف آن¬ها می¬توان به اثرات ضد سرطان، آنتی اکسیدانت و تعدیل کننده سیستم¬ایمنی اشاره کرد. همچنین قارچ ها از منابع پر بازده و کم هزینه در زمینه تولید فراورده های دارویی از قبیل الکل¬ها، استروییدها و منابع غدایی به شمار می آیند. یکی از قارچ¬هایی که مطالعات نسبتا اندکی بر روی آن انجام گرفته Ganoderma resinaceum (Fr).karsten متعلق به تیره Polyporaceae می¬باشد. با توجه به اینکه این قارچ به فراوانی در جنگل¬های استان مازندران یافت می¬شود و قارچ¬های جنس Ganoderma همچون Ganoderma lucidum دارای خواص دارویی و درمانی بسیار بوده و به علت اثرات ضدسرطان مورد توجه قرار گرفته¬اند و با توجه به مطالعات فیتوشیمیایی محدود انجام شده بر روی Ganoderma resinaceum (Fr).karsten ، هدف از انجام این آزمایش بررسی فیتوشیمیایی قارچ جهت شناسایی ترکیبات بیشتر از آن می¬باشد. روش کاراجزای قارچ پس از جمع آوری و خشک شدن توسط دستگاه خرد شده، سپس سه فراکسیون هگزان، اتیل استات و متانول هر بار سه مرتبه به روش ماسراسیون عصاره گیری شدند. عصاره¬ها توسط روتاری در خلا حلال پرانی شدند و هر نوبت پس از TLCگذاری اولیه به ترتیب وارد ستون¬های 5¬سانتی با سیلیکاژل مش درشت و 2¬سانتی با سیلیکاژل مش ریز شدند. ستون با مخلوطی از فاز متحرک شامل هگزان: اتیل استات شستشو شده و فراکسیون¬هایی که الگوهای یکسانی داشتند روی هم ادغام شدند. یافته¬هاچهار ویال به نام¬های E2a، E4c، E4d، E5b از عصاره اتیل استاتی جهت آنالیز H-NMR،C-NMR ، DEPT90 و DEPT135ارسال شدند و پس از بررسی و دریافت نتایج حاصل از طیف و مقایسه با مقالات به ترتیب ترکیبات مشتقات ارگوسترول شامل ارگوسترول پراکساید، 9،11-دهیدروارگوسترول پراکساید، 7،22-ارگوستا¬دی¬ان3-بتا¬ال و ارگوسترول شناسایی شدند.بحثمطالعات فیتوشیمیایی انجام گرفته روی این قارچ طی سال¬های اخیر، منجر به شناخت ترکیباتی از قبیل ارگوسترول، بتاگلوکان، اگزوپلی¬ساکارید، تری¬ترپنویید، بتاسیتوسترول و گانودریک¬اسید شده است. در این مطالعه نیز ترکیبات مشتق ارگوسترول از قارچ مذکور جداسازی شدند.این مشتقات دارای خواص بسیار همچون پیشساز ساخت دارو¬های استروییدی و سنتز ویتامین D2 می¬باشند. با توجه به پر هزینه بودن فرآیند سنتز بسیاری از دارو¬ها می¬توان از ترکیبات قارچی هم بصورت خالص شده و هم بصورت پیشساز استفاده نمود.Mushrooms are natural sources that can be used for anti-cancer, anti-oxidant, and immunomodulatory effects. Also, fungi are considered as suitable sources for the production of pharmaceutical products such as alcohol, steroids and nutrients. One of the fungus found abundantly in the woods of Mazandaran is Ganoderma resinaceum (Fr).Karsten belongs to the family of polyporaceae. According to the limited phytochemical investigations which had done on other species of this jenus, The aim of this investigation is to phytochemical analysis of this fungus.MethodFungal components were crashed by grinder after collection and drying and extracted three times as three fraction of hexane, Ethylacetate, and methanol by maceration method. The extracts were desolvented by rotor-evaporator system and each time after primary TLC were imported to 5cm silica gel high mesh( 0.2-0.5mm) and 2cm silica gel low mesh (0.04-0.06mm) column chromatography respectively. The columns were washed with mixture of mobile phase consisted of hexane: Ethylacetate and subsequently vials with resemble pattern combined with each other and of resulted combined vials, four ones with higher purity and quantity were selected.Findings Three vials of Ethylacetate purified extract called a E2a, E4c, E4d and E5b forwarded for H1-NMR, C13-NMR, DEPT90 and DEPT135 analysis which after analysis and receiving the spectrum results and comparing with articles, respectively ergosterol derived compounds, ergosterol peroxide, 9,11-dehydro ergosterol peroxide, ergosta-7, 22-dien-3β-ol and ergosterol were identified.DiscussionThe fungus Ganoderma resinaceum is of the fungi which there have not been specific attempts on it. Afew phytochemical study had been done to date is for Xian-Qiang Chen et. al 2017 which resulted in identification of Nortriterpenoids compounds which were reported for the first time in this fungus. Other researches on this fungus were correlated to decolorization of methylene blue, degradation of pitch and delignification of wood.ConclusionThree reported compounds in this investigation including ergosterol peroxide, 9,11-dehydro ergosterol peroxide and ergosta-7, 22-dien-3β-ol were reported for the first time in this fungus which all of them were from Ethylacetate extract fraction indicating the grand part of Ethylacetate component are corresponding to steroid and non-polar compounds." @default.
