- W3021366188 abstract "Most experiments in molecular biology involve the use of restriction enzymes at some stage of the experiments. The ability of these enzymes to cut DNA at a specific sequence of bases has greatly stimulated the growth of recombinant DNA technology. This chapter discusses about all commercially available enzymes, their general properties, the basic procedures for their use, and the ways of troubleshooting problems encountered in their use. Most enzymes can be used successfully without the help of premade kits, colorful charts, complex buffers, or specialized equipment. The sequence of the DNA in hand or the design of the experiment may influence the choice of the enzyme. For most base sequences, more than one enzyme is available that recognizes that sequence. An enzyme that generates a four-nucleotide overhang is preferable over its isoschizomer that creates a two-nucleotide overhang if the ends are to be ligated to similarly-generated cohesive ends. In contrast, if the experiment requires ligation of ends generated by enzymes that recognize different sequences, enzymes that generate blunt ends would be preferable over those that create an overhang." @default.
