- W3021514770 abstract "The sensitivity and precision of luminescence assay are limited by many factors. This chapter presents information of a practical nature for the application of luminescence assay in chemical analysis. It has been reported that with most fluorescence spectrometers, the observed excitation and emission spectra contained instrumental artifacts resulting from spectral variations of the components. Instrumental spectra can be converted to true spectra by correction factors derived by suitable calibration methods. It is described that the calibration of a monochromator and photodetector (detector system) for the measurement of the spectral distribution of luminescence requires several steps. The several problems related to luminescence assay discussed in the chapter are problems of light absorption, problems relating to stray light and light scattering, and problem because of photodecomposition. It is also discussed that absorption and emission by the sample cuvette and by the solvent are factors which frequently cause spectral distortion and limit sensitivity." @default.
