- W3028759621 abstract "Objective To construct poly-e-caprolactone (PCL)-gelatin bioengineered patch containing autologous mesenchymal stem cells (MSCs) and explore its effect on the treatment of damaged external anal sphincters (EAS) in rats.Methods Twenty-four Wistar rats which experienced partial sphincterectomy were randomly divided into 3 groups:group A (n =8) subject to EAS injury without treatment ;group B (n =8) subject to surgical repair,and group C (n =8) subject to both surgical repair and application of PCL-gelatin scaffolds previously seeded with autologous MSCs.Functional evaluation of EAS was achieved by anorectal manometry.Histologic evaluation of anal canal and the terminal rectum was done.Results Prior to the graft,the MSC-PLC/gelatin patch was characterized by a preservation of MSC phenotype,a homogeneous cell distribution within the PLC/gelatin scaffold and a low rate of cell growth.In group C,the collagenous fiber which deposited at the sphincter injury site was less than in groups A and B.Increased small dilated blood vessels were seen at the sphincter injury site in group C.Conclusion As an adjunct to conventional surgical repair,MSC-PLC/gelatin patch might favor the healing process via an antifibrotic action and an enhanced tissue perfusion.Key words: Poly-e-caprolactone; Mesenchymal stem cell; External anal sphincter; Fecal incontinence" @default.
