- W3029127047 abstract "ObjectiveTo establish the model of myocardial ischemia-reperfusion injury (MIRI) in rabbit during cardiopulmonary bypass (CPB) and evaluate its effective of insulin resistance (IR) induced by MIRI during CPB.Methods16 New Zealand white rabbits were randomly assigned to two groups. The contol group (n=8) no aortic cross-clamping; the model group (n=8) received aortic cross-clamping for 60 min; and intubation from aortic artery and right atrium to establish CPB. After the aortic opening at 15 min carotid artery blood, coronary sinus blood were collected. moreover, the left ventricular myocardium were harvested, plasma glucose and insulin was detected, then the myocardial net extraction rate of glucose and insulin resistance index (IRI) were calculated. The changes of cardiac hemodynamics were monitored. Finally, the protein expression of Glut4 on myocardial cell membrane were detected by immunohistochemistry.ResultsAfter myocardial ischemia-reperfusion at 15 min, extraction ratio of myocardium glucose was (4.07±2.47)% in model group, which less than control group (26.17±9.78)% (P<0.05). The protein expression of Glut4 on myocardial cell membrane was 85.27±9.23 in model group, which less than control group158.91±13.43 (P<0.05). IRI was 13.65±1.13, while higher than control group 5.38±0.65 (P<0.05).ConclusionA economic rabbit model clinically of myocardial insulin resistance has been successfully established during CPB.Key words: Cardiopulmonary bypass; Myocardial ischemia-reperfusion injury; Insulin resistance; Rabbit" @default.
