- W3030269595 abstract "Objective To investigate the effects of propranolol on the proliferation and apoptosis of the cultured infantile hemangioma-derived endothelial cells (HemECs).Methods The infantile hemangioma tissue harvested from female patients was cultured using the tissue explants adherent method.The tissue was digested to become single cell suspension.The HemECs were labeled and purified by fluorescent-activated cell sorting (FACS).The cell viability was measured by cell counting kit-8 assay (CCK-8).Cell proliferation was studied by calculating BrdU incorporation rate.The induction of apoptosis was determined by flow cytometry analysis of propidium iodide and annexin V-fluorescein isothiocyanate staining.The cells were stained with Hoechst 33342 to determine the percentage of apoptosis.The expressions of VEGF and bFGF were detected by Western blotting.The activities of caspase-3,caspase-8 and caspase-9 were measured by a commercial available caspase activity assay kit.Results Propranolol at a concentration lower than 50μmol/L didn't have any effects on cell viability.However,after treatment with propranolol at 100μmol/L for 24 h,cell viability was significantly reduced to approximately 48％.Meanwhile,the percentage of apoptosis cells was also significantly increased compared with that of the control group (10.7 ± 3.2％ vs.33.5 ± 2.9％,P＜0.01).Furthermore,propranolol treatment suppressed the VEGF and bFGF expressions,and increased the activities of caspase-3 and caspase-9 in HemECs,whereas the activity of caspase-8 remained unchanged.Conclusions Propranolol down-regulates VEGF and bFGF expressions,and increases the activities of caspase-3 and caspase-9,which may be responsible for its ability to inhibit proliferation and induce apoptosis in HemECs.Key words: Hemangioma; Endothelial cells; Propranolol" @default.
