- W3030693463 abstract "Objective To investigate the Smads expression involved in TGF-β1 signal transduction from the transcription level during the development of liver fibrosis in BALB/c mice infected with Schistosoraa japonica and after treatment with interferon-γ( IFN-γ). Methods BALB/c mice, 16th week post infection with cercariae of Schistoma japonicum, were randomly divided into 3 groups: the placebo group, the praziquantel group and the praziquantel combined with IFN-γ group. The treatment lasted for 8 weeks. Liver specimen were harvested at the 8th, 12th, 16th and 24th week, respectively, for the pathologic examination through observing the collagen expression by picfic acid-sirius red staining in the liver tissue section and the mRNA level detection of S mad2,Smad3,Smad4 and Smad7 by RT-PCR. Results Liver fibrosis was established at the 16th week and collagen expression increased along with the time. CoLlagen expression reduced in praziquantel combined with IFN-γ group while it seemed no significant difference between the placebo group and the praziquantel group. During the course of liver fibrogenesis, the level of Smad3 mRNA increased to two fold of the normal level at the 16th week, while the mRNA level of Smad2 decreased at the 12th week and returned to the normal level at the 16th week. No significant difference was observed in Smad4 and Smad7 mRNA. After combination treatment, Smad7 mRNA elevated significantly, Smad2 mRNA remained at low level and Smad3 mRNA was higher than that in the normal control group. No significant difference was found in Smad4 mRNA level. Con-clusion Up-regulation of Smad3 mRNA, down-regulation of Smad2 mRNA and constant level of Smad7 mRNA may lead to live fibrogenesis in mice. IFN-γ may induce the increase of Smad7 mRNA to inhibit liver fibrosis.Key words: Smad protein; Liver fibrosis; TGF-β1; Schistosomiasis" @default.
