- W3030699443 abstract "Objective To detect the cytoskeleton change of pancreatic epithelial cells after oxidative stress stimulation,and investigate the mechanism of the structure change in three-dimensional culture system.Methods Pancreatic epithelial cells were cultured in matrigel to construct a three-dimensional culture model.The cultured cells were stimulated with 1 or 200 μmol/L H2O2 for 15 min,and then the cell morphology changes were observed after 15 min,1 h and 4 h.The cytoskeleton of these cells were stained immunofluorescently and observed under a laser confocal microscope.Enzyme linked immunosorbent assay (ELISA) was used to detecte the nuclear content of nuclear factor-κB (NF-κB).Results 200 μ mol/L H2O2 could induce cell contraction after 15 min.One h later the cell morphology recovered,but 4 h later,cells shrinked more apparently.Intracellular actin and microtubules lost rapidly after H2O2 treatment,but returned to normal structure after 1 h.Four h later,cytoskeleton became fuzzy and eventually disintegrated.1 μmol/L H2O2 did not induce any effect on the morphology and cytoskeleton of pancreatic epithelial cells.200 μmol/L hydrogen peroxide treatment increased the activity of NF-κB gradually,and 1 μmol/L hydrogen peroxide treatment had little impact on the activity of NF-κB.Conclusion Oxidative stress can induce early onset of reversible cell contraction and cytoskeleton depolarization in pancreatic epithelial cells,and the increase of NF-κB expression.Key words: Pancreatic epithelial cells; Hydrogen peroxide; Cytoskeleton" @default.
