- W3030931345 abstract "ObjectiveTo investigate the influence of Wnt inhibitor XAV939 on apoptosis of gastic cancer MKN-45 cells.MethodsMKN-45 cells were cultured with different concentrations of XAV939. The inhibition of cell proliferation was detected by methyl thiazol tetrazolium (MTT), and cell apoptosis was tested by Annexin V-fluoresceine isothiocyanate (FITC)/propidium iodide (PI) double staining analysis and TdT-mediated dUTP nick end labeling (TUNEL) labeling method. Annexin V-FITC/PI was used to detect the early apoptotic cells. The expression levels of c-Myc, B cell lymphoma/leukemia-2 (bcl-2), and β-catenin mRNA were detected by real-time quantitative polymerase chain reaction (Real-time PCR), and the protein expression of poly adenosine diphosphate-ribose polymerase (PARP), c-Myc, bcl-2, bcl-2 associated X protein (bax), Axin 1, β-catenin, phosphorylated β-catenin (p-β-catenin) and Cyclin D1 by Westem blotting.ResultsInhibitory rate of MKN-45 cells treated with XAV939 for 48 h was gradually increased. Early apoptotic cells were detected by Annexin V-FITC/PI double staining analysis and TUNEL labeling method. After treatment with XAV938 for 12 h, the apoptosis rate of MKN-45 cells was 8.8%, 13.5%, 21.5% and 25.8%, respectively. After treatment with different concentrations of XAV938 for 48 h, the mRNA expression levels of c-Myc, bcl-2 and β-catenin were decreased in a concentration-dependent manner (1.0, 2.0, 4.0 μmol/L) as compared with control group, meanwhile, protein expression levels of c-Myc, bcl-2, PARP (116×103), β-catenin and Cyclin D1 were down-regulated, and those of bax, Axin 1, p-β-catenin and PARP (85×103) were up-regulated.ConclusionXAV939 can promote apoptosis of MKN-45 cells, which may be associated with down-regulation of Wnt/β-catenin signaling pathway.Key words: XAV939; Gastric carcinoma; Wnt/β-catenin signaling pathway" @default.
