- W3031004027 abstract "ObjectiveTo investigate the expression of peroxisome proliferator-activated receptorγcoactivator-1α(PGC-1α)and the genes associated with mitochondrial synthesis in the peripheral skeletal muscle of chronic obstructive pulmonary disease of(COPD) rat model.MethodsThe tumor necrosis factor-α (TNF-α) were detected in blood and skeletal muscle respectively, the PGC-1α,nuclear respiratory factor 1 (NRF1), mitochondrial transcription factor A(Tfam), cytochrome coxidaseⅣ(COX)ⅣmRNA were detected by real-time fluorescent quantitative polymerase chain reaction(FQ-PCR), the relative content of PGC-1α, NRF1, Tfam,COXⅣprotein were detected by Western blotting method in the control group and the COPD group.ResultsThe TNF-αwas significant increased in peripheral blood and skeletal muscle in COPD model rats(ng/L, 378. 09±26. 34, 330. 56±23. 79 vs.154. 70±12. 98, 129. 77±11. 79), and the mRNA and protein of PGC-1α(0. 17±0. 05 vs.1. 00±0. 00), NRF1(0. 20±0. 08 vs.1. 00±0. 00), Tfam (0. 21±0. 04 vs.1. 00±0. 00), COXⅣmRNA(0. 25±0. 07 vs.1. 00±0. 00)were significantly reduced in COPD model rats.ConclusionTNF-αmay beinhabited PGC-1αand mitochondrial synthesis-related gene expression in skeletal muscle of COPD rat.Key words: Chronic obstructive pulmonary disease; Mitochondria; Peroxisome proliferator-activated receptorγcoactivator-1α" @default.
