Finally, we efficiently purified a new Δ-9-desaturase KRICT Rt9 protein from the transgenic E. coli . The purified enzyme Δ-9-desaturase KRICT Rt9 was reacted with Stearoyl-CoA and NADH to produce oleic acid in vitro . • Codon-optimized gene was expressed in E. coli and purified as a new Δ-9-desaturase. • The amount of oleic acid was increased by greater than 3.25-folds in vivo . • The oleic acid was produced in vitro by Δ-9-desaturase with Stearoyl-CoA and NADH. • This is the first report to make an unsaturated fatty acid enzymatically in vitro ." @default.
