- W3036966506 abstract "Controversy exists as to whether tissue glycogen is preserved or destroyed by treatment with uranyl acetate before dehydration. Some authors report that en bloc staining with uranyl acetate destroys glycogen ( 1 ), whereas others show preservation of glycogen following this staining procedure ( 2 ). Our study on conducting fibers of the dog heart suggests that there might be another factor involved in the preservation of several cytoplasmic structures which is vital to the understanding of glycogen preservation after the treatment with uranyl acetate. Fibers of conducting tissue are composed of a specific type of muscle cell containing large cytoplasmic areas filled with glycogen. Present observations were focused on these areas. Material was fixed in 3%. glutaraldehyde in phosphate buffer pH 7.2, post-fixed in 0 S O 4 and processed without (control) or with uranyl acetate." @default.
