With recent advancements, there has been significant rise in drug molecules, both small molecule drugs and large biopharmaceuticals, which need an aid from formulation scientists to achieve the desired therapeutic efficacy. For example, nanoparticles have been used to enhance the solubility of small molecular drugs, and to protect and increase the intestinal permeability of protein/peptide drugs, which in turn enhances the in vivo performance of the encapsulated drug molecule. This trend has resulted in increased prominence of nanoparticles based oral delivery systems. During the preclinical development, the animal experimentations are an important part for evaluation of new formulations. The complex nature of in vivo studies requires utmost care when designing the experiments, selecting the appropriate animal model and techniques to generate reliable results. This book chapter highlights and discusses the overview of rationale and challenges of in vivo studies, the animal models and in vivo techniques for oral delivery of nanoparticles. A wide range of factors and methods are discussed in this chapter including pharmacodynamics and pharmacokinetic profiling, imaging methods, organ/tissue analysis and histology." @default.
