- W3048567238 abstract "I explore the possibility of detecting singlenanoparticles below 100 nm in real time. The motivation behind thisproject is the development of an optical sensor capable ofmonitoring the environment for harmful viruses or other nanoscopicpathogens. The main difficulty in detecting small particles arisesfrom a strong signal dependence on particle size. In a standardapproach, it is customary to measure the intensity of lightscattered by a particle. This measurement has R⁶ dependence onparticle size, due to which the signal falls quickly below thenoise level when attempting to detect smaller particles. The limitof currently available single particle detectors is approximately200 nm for transparent glass-like particles. On the other hand, myapproach investigates two different optical techniques, both ofwhich employ a weaker particle size dependence. The weaker sizedependence in our schemes leads to a better signal-to-noise ratiofor small particles. The first method is basedon measuring the optical gradient force, which acts on the particleinside a strongly focused laser beam. The optical force isproportional to the third power of particle size, R³. Similar tomass spectrometry, the laser focus influences the trajectory of aparticle as it passes through a tightly-focused laser beam.Particle trajectory is monitored using quadrant detector placed intransmission. I successfully detect and recognize R = 50 nm and R =100 nm particles in a mix. I show that such force measurement isnot background-free due to the Brownian motion. I demonstrate theways to minimize the contribution of the Brownian motion, anddiscuss the limitations of the scheme. Theother detection approach is based on the interferometric detectionof the scattered light amplitude. The approach represents aninterferometer, where the scattered light forms one arm. Thescattered light and the reference beam are combined on a splitphotodetector, which renders a background free signal. I analyzesignal-to-noise, and demonstrate real-time detection (with timeresolution of 1 ms) of particles down to R = 7 nm. In the finalstage of this work, I apply this method to detecting live viruses.I successfully detect and distinguish Influenza and Sindbis virusesfrom a mix of polystyrene particles of similarsize." @default.
