- W307251710 abstract "In prototroph M-506, the genes coding for antimycin A (ant) and melanin (mel) formation were resistant to plasmid “curing” treatments: these genes would be integrated in the chromosome. In all the auxotrophs descended from M-506, ant and mel can exist autonomously as extrachromosomal elements. They tend to be reintegrated into the chromosome of prototrophic revertants. This episomal movement in and out of the chromosome could occur by translocation mechanisms. The gene coding for aerial mycelium formation (amy) was stable to “curing” in prototroph M-506, but extremely unstable in auxotrophs and prototrophic revertants; in the latter strains, amy would be located on a plasmid; it does not behave as an episome. Since they are generally “cured” together, ant and mel would code for some common functions probably involving the bioconversion of tryptophan into the salicylic acid moiety of antimycin A and of tyrosine into melanin. Plasmid DNA could be isolated from Ant+ auxotrophs, not from Ant+ prototroph nor from Ant– strains; however, its biological activity could not be demonstrated.
