- W308517564 abstract "Publisher Summary This chapter summarizes the findings of herpes simplex virus (HSV)-induced transformation i n vitro, contrasts the finding with transformation induced by other DNA viruses, and considers mechanisms which may be operative in the HSV system. Transformation refers to the spectrum of phenotypic changes, which the mammalian cells may undergo either spontaneously or following treatment with one or more “transforming agents.” Cells non-permissive for a DNA virus do not produce infectious virus, although the cells may undergo a lethal infection depending on the stage during the virus cycle when the infection is aborted. The mutant virus must be able to infect the resistant cells, and the productive virus cycle must be aborted at a stage early enough to avoid cell death. Malignancy in vivo or tumorigenic transformation in vitro is a multistep process. HSV transformed cells have been identified morphologically on the basis of their appearance compared to surrounding “non-transformed” cells. Morphological and/or tumorigenic transformation by HSV is a relatively inefficient process. Expression of viral proteins in HSV-transformed cells has been detected primarily by immunofluorescence. In situ hybridization has been employed for detecting viral RNA in transformed cells. The ability of HSV to damage chromosomes and induce reparable DNA damage is consistent with the properties of carcinogenic and/or mutagenic agents." @default.
