- W3094038923 abstract "The HPV oncoproteins, E6 and E7, are expressed in cervical cancers and have the potential to serve as unique tumor specific antigens. We sought evidence for immunoreactivity to these viral proteins in cervical cancers from patients receiving radiation therapy. Cervical tumor brushings of 37 gynecologic cancer patients enrolled on a prospective trial were collected at baseline, during chemoradiation (weeks 1, 3, 5, and 3-month follow-ups). Cervical tumor brushings were collected from exophytic cervical cancers and incubated with peptides spanning the HPV 16 E6 and E7 proteins. Cells were analyzed using multispectral flow cytometry to identify CD8+ T cells expressing the activation marker, IFN-gamma, following treatment with HPV peptides or PMA control. Mean percentages were compared with an unpaired t-test. The mean percentage of CD8+ effector T-cells expressing IFN gamma at baseline, week 1, week 3, week 5 and, 3 months follow up of radiation therapy was 2.4%, 0.2%, 1.5%, 1.6% and 5.2%. The percentage of patients with at least 1% HPV reactive CD8+ T-cells at each time point was 20% (6/30), 5% (1/20), 32% (6/19), 28% (7/25), and, 10% (1/10). By comparison, there was no change in the percentage of T-cells expressing IFN-gamma at each time point when brushings were treated with control PMA. Radiation therapy may result in generation or expansion of HPV reactive T-cells within cervical cancers. Initial declines in HPV reactive T-cells in the first week of treatment were followed by expansion in the third and fifth week of radiation treatment. Further investigations will be needed to determine if the presence of intratumoral HPV effector T-cells is associated with favorable outcomes following radiation therapy." @default.
