Initially recognized as Ca2+-dependent receptors binding carbohydrates via one or more C-type lectin-like domains (CTLD), their ligand repertoire was quickly found to include not only carbohydrates but also proteins, lipids, crystals or inorganic molecules dependent or independent of Ca2+. In the immune system, the myeloid C-type lectin receptors (CLRs) play a crucial role either triggering, modulating or inhibiting a variety of effector functions (phagocytosis, antigen presentation, cytokine production) often categorized by their ligands, localization and downstream signaling. They are mainly expressed by antigen-presenting cells (APCs) and contribute to the recognition of pathogens (fungi, bacteria, viruses and parasites) but may also bind to self-antigens and endogenous danger signals released by damaged or necrotic cells to contribute to homeostasis and maintain self-tolerance. In this article, we will systematically discuss major superfamily members. Specifically, myeloid CLRs will be highlighted that play an important role in diverse physiological processes including homeostasis, pathogen control, and autoimmunity." @default.
