- W3119646145 abstract "Purpose: To further develop the application of the GoldenGate Assay for high-throughput screening of up to 1536 somatic mutations in clinical cancer specimens. Experimental Procedures: Assays for 192 somatic mutations were selected from an initial set of over 300 assays targeting mutant alleles in 40 genes frequently mutated in a variety of tumor types. Pancreatic cancer samples collected from tumor tissue, serum, and stool were used to test these assays. Normal human Coriell DNA samples were also screened to obtain baseline intensity data for background normalization of the clinical samples. All samples were quantified by PicoGreen, whole-genome amplified, and screened with the assay pool. To test assay sensitivity, oligos each containing one of 31 different mutations in TP53 and Kras were also spiked into a normal sample and screened in parallel. Finally, samples with high relative mutant signal intensities were mixed with normal samples in varying amounts to further test assay sensitivity for additional targets. Results: Separating assays for mutant alleles into multiple pools resulted in improved assay performance in detecting both the normal alleles and spiked-in mutant alleles.
