- W3130070654 abstract "Abstract Safflower (Carthamus tinctorius L.) is an important oilseed crop but a high yield loss is due to fungal disease Alternaria carthami. The study was aimed at the development and refinement of plant regeneration protocols for obtaining a high frequency of shoot regeneration and the tissue culture technique has been extended for production of plants resistant to A. carthami. A highly efficient protocol is developed for indirect or direct organogenesis from different explants in solid culture with Agrobacterium strain LBA 4404 containing a rice chitinase (Chi 11) gene. Different culture mediums were tested in addition to inorganic and organic nitrogen sources and also combination of amino acids into the reference medium. Safflower is amenable to manipulations in vitro and genetic transformation protocols through vector-mediated gene transfer are in place providing scope for development of transgenics for desirable traits. In our study, high frequency of shoot elongation was observed in MS medium supplemented with low level of BA and GA3, but further increase in plant growth regulators concentration promoted callusing. All the transgenic shoot tip explants cultured on medium fortified with TDZ (3.5 μM) and NAA (1.5 μM) along with hygromycin showed highest percentage of shoot induction (96%) and maximum of 32 average number of multiple shoots were formed. However, in our observations, shoots cultured on NAA and AgNO3 supplemented medium induced efficient rooting. The reduced concentration of salts, sucrose could be supportive to promote rooting in NARI-6. Micropropagation of safflower provides an opportunity to harvest virus-free material and conserves important germplasm resources in vitro. It furnishes material for genetic transformation and for employing molecular techniques inbreeding." @default.
