- W3131405320 abstract "Long QT syndrome (LQTS) is an inherited ventricular repolarization disorder. Patients with the QT interval prolonged might develop lethal ventricular arrhythmias such as Torsade des pointes and sudden cardiac death. There are two major currents involved in the ventricular repolarization, IKr and IKs currents. Loss-of-function mutations on any of the genes that encode the proteins involved in the generation of these two currents are directly related to LQTS. IKs channel is formed by four alpha subunits, coded by the KCNQ1 gene, co-assembled with regulatory beta subunits, encoded by the KCNE1 gene. Mutations in these two genes generate LQT1 and LQT5, respectively. Defects in pore-forming subunits of hERG, which suppress IKr current, lead to LQT2. Polyunsaturated fatty acids (PUFAs) have been described as modulators of voltage-gated ion channels. They produce a wide range of effects depending on their nature (e.g., ω-3/6/9) and on the kind of channel affected. PUFAs, which specifically inhibit NaV and CaV, and those that activate KV currents, are considered promising anti-arrhythmic drugs. In the present study, we use high-resolution all-optical electrophysiology on ex-vivo Zebrafish (ZF) heart to assess the anti-arrhythmic properties of PUFAs. To achieve an LQTS phenotype, ZF hearts were incubated with different IKr and IKs blockers (E4031, Chromanol 293B, and JNJ303). PUFAs and PUFA analogues were then applied to restore the wild-type phenotype of the ZF heart. Moreover, ZF hearts were incubated with Isoproterenol to stimulate the sympathetic cardiac response, in which IKs currents play a more important role in the ventricular repolarization. Ventricular action potential duration (APD) and ventricular calcium transients were then analyzed. PUFA analogues showed a tendency of APD shortening on the ZF hearts. This result suggest that PUFAs could have a therapeutic potential for the treatment of LQTS." @default.
