- W3131567203 abstract "High-speed atomic force microscopy (HS-AFM) is a powerful technique that provides dynamic movies of biomolecules at work. HS-AFM has several strengths that make it particularly powerful for the study of protein complexes involved in membrane deformation and trafficking: (i) HS-AFM operates at ambient temperature and pressure, (ii) images unlabeled biomolecules, (iii) achieves ∼1nm lateral, ∼0.1nm vertical and sub-second temporal resolution, (iv) allows the exchange of environmental buffer conditions, and (v) allows the mechanical manipulation of the protein complexes under investigation. Many of the membrane-associated proteins involved in membrane deformation and trafficking form supramolecular complexes that are non-regular and adapt to the changing membrane topology, eg. ESCRT-III spirals, Clathrin coats. The high signal-to-noise ratio in HS-AFM movies allows to directly image such individual supramolecular structures without the need of averaging methods. Here, we will present our most recent technical and application progresses using HS-AFM for the analysis of membrane deformation complexes, and discuss how these supramolecular structures can provide the considerable energetics necessary for membrane deformation processes." @default.
