- W3132899686 abstract "Natural killer T (NKT) cells are a subset of T cells, which play key roles in the interface between adaptive and innate immunity. The combined efforts of scientists in diverse fields have led to discoveries of the types of antigens recognized by NKT cells, and these include a diverse collection of glycolipids. The potential impacts of stimulation of NKT cells by these antigens has led to efforts to optimize antigen structures to generate desired responses from NKT cells. These structure-activity studies have resulted in development of glycolipids that show promise in multiple pre-clinical and clinical studies, including uses as vaccine adjuvants and in cancer immunotherapy. The conservation of NKT cells among most mammals led to speculation that this cell type is involved in surveillance for exogenous antigens signaling infection, and glycolipids from multiple pathogens have been discovered that are antigens for NKT cells. Early in the study of NKT cells, autoreactivity was observed between NKT cells and antigen-presenting cells. This observation suggested that there are endogenous antigens produced that stimulate NKT cells, and multiple proposed glycolipids have been proposed as these antigens. This article describes the outputs of efforts to optimize glycolipid structures for NKT cell stimulation, identification of exogenous antigens for this cell type, and proposed structures of endogenous antigens." @default.
