- W313808423 abstract "Publisher Summary Synthetic glycoconjugates, materials that are often referred to as neoglycoconjugates, were first prepared some 70 years ago as products of an intellectual exercise. The neoglycoconjugates made were not sophisticated from the point of view of carbohydrate chemistry, as they involved only simple mono- and di-saccharides. However, with the recognition of the importance of these substances, new conjugation protocols have been developed and, more often than not, the carbohydrates to be conjugated included complex oligosaccharides resulting from intricate chemical syntheses. As such, these materials are generally labor-intensive and, therefore, expensive commodities. The actual expense, and the wasteful nature of the conjugation reaction—as it is often performed—is multiplied by the fact that, in order to maintain a reasonable reaction rate, conjugation of carbohydrates to proteins is normally conducted at a high molar hapten–carrier ratio. When the reaction is terminated, the only material isolated from the conjugation mixture is usually the neoglycoconjugate, even though the mixture may still contain unused hapten. Therefore, when expensive materials are converted into neoglycoconjugates, recovery of unused ligand from the conjugation mixtures ought to be considered. The protocols for recovery of excess hapten described in this chapter have resulted from author's experience in conjugating carbohydrates to proteins, using squaric acid diesters. The procedures are simple and can be readily included in overall protocols, involving other conjugation chemistry, provided a large part of the hapten used in excess remains unchanged." @default.
