- W3138129438 abstract "Natural products of plants are traditionally representing a source for bioactive substances. Boswellia sacra (BS) resin has been commonly used in traditional medicine in Arab countries for its analgesic, anti-microbial, and anti-inflammatory properties, which reflect their potential as immunomodulatory agent. Dendritic cells (DCs) are professional antigen-presenting cells (APCs) and sentinel cells that regulate the immune response. It has been proven that several herbs and plant extracts, crude and purified, have the efficacy to modulate immune response including DCs. Aim: The aim of this study is to investigate whether crude Boswellia sacra essential oil (BSEO), from Sultanate of Oman, has a potential effect on the phenotype and function of human monocyte-derived DCs. Methods: Oil extract of Boswellia sacra was prepared by hydrodistillation using a custom made hydrodistiller. BSEO-mediated cell viability has been initially studied on human skin dermis cells (HSD) and DC precursors using quantitative and qualitative assays before applying on DCs. Then, human DCs were generated and differentiated from peripheral blood monocytes in the presence of granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin 4 (IL-4). DCs have been exposed to 5 µg/mL or 10 µg/mL of BSEO in vitro. Morphological, phenotypical and functional properties have been studied by microscopy, flow cytometry, and ELISA. Results: BSEO was found to interfere with DCs differentiation from precursor cells, unable to induce full DC maturation, and suppress DC maturation in the presence of LPS. In addition, endocytic capacity of DCs treated with BSEO was enhanced in comparison to LPS treated cells. Treatment of DCs with 5 µg/mL of BSEO resulted in an increased production of IL-10 and increased proliferation of allogeneic T cells. Conclusions: Our data indicated that BSEO deviates differentiation of monocytes away from mature DCs toward tolerogenic DCs. These findings can potentially be employed to generate tolerogenic DCs that are able to induce tolerance in hypersensitivity, autoimmunity as well as transplantation." @default.
