- W314221972 abstract "Publisher Summary This chapter highlights the problems concerning steroid radioimmunoassay (RIA). The main problem in steroid RIA is the obtention of antisera specific enough to discriminate among closely related structures. Depending on the specificity of the antiserum obtained, a more or less sophisticated purification procedure is applied to the sample prior to the assay. Any separation technique involves the problem of reagent blank, the extent of which, in turn, depends on the affinity of the antiserum. Thus, as is usual in RIA, the problem of setting up well standardized assay procedures can be ultimately reduced to the preparation of suitable antisera. A minor problem in steroid RIA is related to the tracer used. Because of the relatively low affinity constants of the antisera, steroid RIA requires a careful optimization of the parameters governing the assay. The conditions to be fulfilled for steroid RIA are the following: effective steroid separation, low reagent blank, high chemical recovery, and possibly a simple methodology capable of being standardized. The 125I-labeled cortisol-21-TME derivative was found to be a perfectly suitable tracer for competitive protein binding (CPB) analysis of plasma cortisol, although a slightly lower affinity with transcortin resulted as compared with tritiated cortisol. The stability on storage of 125I-labelled testosterone and cortisol, and aldosterone in ethanol solution was checked for periods up to 60 days; no significant deiodination was observed by paper electrophoresis and thin layer chromatography and no degradation trend was indicated by the dose-response curves obtained." @default.
