Such bioassays rely on the receptor–reporter principle, wherecell lines are transfected with plasmids carrying a reporter gene undercontrol of environment-dependent promoters (receptor), whichplay a key role in regulating gene expressions in response to extracellularsignals. We developed the recombinant human lung epithelialcell line A549-NF-κB-EGFP/Neo carrying a genetically encodedfluorescent indicator for monitoring activation of the NF-κB signalingpathway in living cells in response to genotoxic and cytotoxic environmentalinfluences. With this cell line we screened several candidatehuman radiation-responsive genes (GADD45β, CDKN1A) andNF-κB-dependent genes (IL-6, NFκBIA, and pNF-κB-EGFP) for geneexpression changes by quantitative reverse transcriptase polymerasechain reaction (qRT-PCR) assay, using cDNA obtained from total RNAisolated at various time points after exposure to combustion generatednano-sized particle samples." @default.
