- W3146022083 abstract "We have studied the fluorescence decay kinetics of apoand holopyruvate decarboxylase from brewer's yeast (APDC and HPDC respectively) in the native state as well as after the de~turation by 6 M urea and heating of the solution up to 40 C. Statistical analysis has shown that the fluorescence decay function with good fitting criteria my be represented as a sum discrete of three exponential terms or two continuous distribution (Gaussian distribution) of the lifetimes. In this connection a question arises as to which of the models considered in the most adequate one. Pyruvate decarboxylase is a complex protein containing more than 20 tryptophane residues which determine its intrinsic fluorescence with a maximum at 330 nm. Therefore it is unlikely to devide all protein tryptophanyls into 3 classes wi th similar values of lifetimes. We think that in this case only the representation of fluorescence decay function as a continuous distribution of the lifetimes has a physical meaning. For APCD, this simulation includes one wide distribution with center at about 4 ns and a narrow one with the center at about 1 ns (fig.1,a). It is evident that the type of these distributions is determined by the joint action of dynamic and sterical factors, i. e. the conformational of tryptophane residues relative to the polypeptide chain. The majority of tryptophanyls forming the first distribution (Group A) show relatively low values of the fluorescence quenching rate constant which, however, vary considerably due to mobility and fluctuations of the protein structure. Some tryptophanyls within APDC demonstrate high values of the quenching rate constant and form second group chromophores (Group B). The formation of the enzyme's holoform is accompanied by considerable changes in the deconvolution parameters. On the one hand, the distribution A is shifted towards greater" @default.
