- W3146023906 abstract "Very recently, our task group has been the pioneer in finding (pro) renin receptor [(P)RR] expression in human umbilical vein endothelial cells (HUVECs). So, it is important to determine whether prorenin leads to endothelial abnormalities through (P)RR independent on angiotensin II, and whether HRP exists effect on (P)RR blocking. We cultured the primary HUVECs in vitro. Cells' AT1/AT2 receptors of angiotensin II were blocked by olmesartan and PD123319, respectively. Meanwhile, the (P)RR of HUVECs was interfered with small inhibiting (siRNA) and treated with HRP, respectively. PI3K/AKT signaling pathway was blocked with Wortmannin. Then, the treated HUVECs were stimulated with renin and prorenin. The efficiency of (P)RR interfering was detected by reverse transcriptase and PCR (RTPCR). Phosphoinositide3-kinase (PI3K), serine/threonine protein kinase (AKT), phospho-AKT (p-AKT) and nuclear transcription factor-P65 (NFkB-P65) were evaluated by western blot. The concentration of interleukin-6 (IL-6) in cultural medium was detected by enzyme-linked immunosorbent assay (ELISA) kit. Our study showed that the expressions of PI3K, p-AKT, p65, and IL-6 were up-regulated by renin and prorenin. Such effect could be inhibited by (P)RR–RNA interference, but not by HRP. The activations of P65 and IL-6 were not be inhibited by Wortmannin. We found that both prorenin and renin can activate the expressions of PI3K, p-AKT, P65 and IL-6 through (P) RR, and independently of Ang II generation. HRP does not have effect on (P)RR blocking. The expressions of P65 and IL-6 may not be regulated through PI3KAKT signaling pathway." @default.
