- W3147058438 abstract "There is an increasing need for practical, efficient, and inexpensive ways to explore inherited mutations responsible for genetic diseases, acquired mutations involved in cancer development, and induced mutations in mutational spectrometry. Most of the current methods used for mutation screening are based on PCR, and this has solved the problem of target limitation. Simple and efficient methods such as allele-specific hybridization, allele-specific amplification, ligation, primer extension and artificial introduction of restriction sites, and variants of these methods have been developed for screening large numbers of samples for one particular mutation (for review see Cotton, 1993). In many instances, however, screening for unknown mutations is required. Sequencing represents a reliable mutational screening technique, although PCR and direct sequencing is not very sensitive in detecting mutations present in only a fraction of the cells examined, which is often the case in tumor specimens. Sequencing is also very labor intensive unless expensive highly automated sequencers are applied. The development of simpler and less expensive screening techniques like single-strand conformation polymorphism (SSCP) and denaturing gradient gel electrophoresis (DGGE) has made it possible to screen larger series for mutations in several different genes. Nevertheless, improved screening techniques are still needed. Constant denaturant gel electrophoresis (CDGE), which is a modification of the conventional parallel DGGE (Hovig et at., 1991), represents, in my opinion, such an improvement. DGGE was first described by Fischer and Lerman (1983) and is discussed in detail by Fodde and Losekoot (this volume). The separation principle of DGGE and CDGE is based on the melting behavior of the DNA double helix of a given fragment. This melting behavior is sequence dependent, and occurs at domains, rather than at single bases. The melting is detected as a reduction in the mobility ofthe DNA fragment as it moves throughout an acrylarnide gel as a consequence of partial strand" @default.
