- W3171868896 abstract "SUMMARY This chapter reviews the demonstration of the analgesic and anti-inflammatory effects of phenylbutazone and indomethacin, and attempts to explain their mode of action in terms of effects on mediators of inflammation, tissue and cellular metabolism and reaction with protein molecules. Human and animal studies reveal very different half-lives and metabolic pathways and these factors have important clinical implications. A short survey of experiences indicates their points of value in the commoner rheumatic conditions. Intolerance and adverse reactions are detailed, together with some indications of the high-risk patients and the methods for minimising these reactions.
