- W3173487006 abstract "Cytokines participate in inflammatory processes within the kidney, but their effect on renal microvascular function is unclear. We tested the hypothesis that IL-6 attenuates afferent arteriolar autoregulatory responses, and vasoconstriction by ATP and adenosine, using the juxtamedullary nephron technique in wild type (WT; C57/BL6J) and IL-6 knockout mice (IL-6 KO), respectively. Kidney perfusion began at 100mmHg and was adjusted to 80, 100 120 and 140mmHg for determination of autoregulatory responses. Baseline arteriolar diameters were similar and averaged 21.1±0.6 and 21.4±0.4μm in WT and IL-6 KO mice; respectively. WT mice did not exhibit a significant pressure dependent vasoconstriction as arteriolar diameter declined by 3±2% when perfusion pressure was increased from 80 to 140mmHg. In contrast, arteriolar diameter declined significantly by 11±4% (P<0.05) over the same pressure range in IL-6 KO kidneys. ATP mediated vasoconstriction of afferent arterioles is enhanced in IL-6 KO kidneys (n=5). ATP (0.1, 1 and 10μM) reduced afferent diameter by 2±1, 6±3 and 12±3%, respectively, whereas diameter declined by just 1±2, 2±2 and 3±1%, respectively in WT kidneys. Adenosine mediated vasoconstriction of afferent arterioles was also enhanced in IL-6 KO kidneys. Adenosine (0.1, 1 and 10μM) mediated vasoconstriction averaged 0±1, 1±5 and −2±2%, respectively in WT kidneys (n=4) compared to −1±4, 4±4 and 7±3% in IL-6 KO kidneys (n=4). These data suggest that IL-6 attenuates renal microvascular autoregulatory responses and that this attenuation could be through reduced responsiveness to purinoceptor activation.
