- W3176448438 abstract "ザンドホッフ病（SD）は，HEXB遺伝子の変異によりリソソーム酵素であるβヘキソサミニダーゼの機能低下が起こり，その基質であるGM2ガングリオシドなどが分解されずに蓄積してしまう疾患である．主として神経系にGM2ガングリオシドなどが蓄積するために，進行性の神経障害を起こす．その症状は，運動障害や振戦，視覚異常，精神運動発達の遅延や退行などの神経系障害の症状を呈するが，そのメカニズムは不明な点が多く残っている．最も重篤な症状は，古典型で3歳から5歳の幼児期に死亡する．SDモデル動物であるHexb遺伝子欠損マウス（Hexb－/－）は，古典型にみられる症状を示す．私たちの解析した結果では，Hexb遺伝子欠損マウス脳において，運動機能の低下が見られる時期より，かなり早期からミクログリアやアストロサイトの活性化が観察されることが見つかった．早期からのミクログリアなど免疫系細胞の活性化を抑制することで，アストロサイトの活性化を抑制できること，さらに運動機能の低下が改善することを見いだした．また，Hexb遺伝子欠損マウス脳において，脳炎症が進行している時期のアストロサイトにアデノシンA2A受容体が発現し，これを抑制することで炎症性サイトカインやケモカインの発現を抑制し，ミクログリアの活性化を抑制することができ運動機能の低下が改善することを見いだした．本稿では，私達の研究室でHexb遺伝子欠損マウスの解析により見つかってきた異常と新規治療標的としてのグリア細胞について紹介する．" @default.
