- W3176764685 abstract "Immunity is inextricably linked to cancer outcomes, facilitating tumor initiation and progression or eradication. This phenomenon is exemplified by the role of macrophages, archetypical phagocytes functioning on the boundary between innate and adaptive immunity. Tasked with tissue development, restructuring, and maintenance during homeostasis, such functionality is quickly subverted within the inhospitable tumor-microenvironment. Tumor-associated macrophages facilitate neoplastic transformation, tumor immune evasion, and the subsequent metastatic cascade. Herein, we discuss the dynamic process whereby molecular and cellular features of the tumor microenvironment act to license tissue repair mechanisms of macrophages, fostering angiogenesis, metastasis and the support of cancer stem cells. In this article we illustrate how tumors induce, then exploit trophic macrophages to subvert innate and adaptive immune responses capable of destroying malignant cells. We examine sex-based and racial differences in macrophage development and functionality that impact disease course and cancer therapy. Finally, we discuss compelling evidence from murine models of cancer and clinical trials in support of macrophage-targeted intervention either independently or in conjunction with conventional strategies highlighting the potential to dramatically reduce cancer morbidity and mortality." @default.
