- W3178236885 abstract "The NAD + -dependent deacetylases, sirtuins (SIRTs), regulate mitochondrial biogenesis via deacetylating PGC-1α, whereas the protein acetyltransferase, GCN5, inhibits PGC-1α nuclear translocation by its acetylation. NADase CD38 competes with SIRTs for cellular NAD+ pool. High-fat diet activates CD38 and/or GCN5 while downregulating NAMPT, the rate-limiting enzyme in NAD salvage pathway, thus decreasing cellular NAD+ level and SIRTs activity. PURPOSE: To investigate the effects of 12 wks of EVOO supplementation alone and in conjunction with endurance training on muscle levels of SIRTs and NAD + -dependent enzymes, as well as mitochondrial biogenesis in rats fed a high-fat diet. METHODS: Female Sprague-Dawley rats, aged 4-6 wks, were divided into 4 groups: 1) chow diet (C, N = 6); 2) fed a high-fat/high-cholesterol atherogenic diet (Ath, N = 12); 3) fed the Ath diet containing 20% EVOO (EVOO, N = 12); and 4) fed Ath + EVOO diet and trained (25 m/min, 10% grade for 60 min/day, for 12 wks; EVOO+T, N = 12). RESULTS: Total protein acetylation was higher in Ath vs. C, EVOO, and EVOO+T (P < .05), respectively. Ath and EVOO increased levels of GCN5 and CD38, whereas the effect was attenuated with EVOO+T (p < .05). EVOO+T elevated NAMPT level vs. C (P < .05). SIRT1 was increased by ~1.8-fold (P < .05) in Ath, EVOO, and EVOO+T compared with C, whereas SIRT3 was not affected. SIRT6 content was also increased in EVOO (P < .05) and EVOO+T (P < .01) vs. C. PGC-1α was not affected by Ath, but was upregulated by EVOO and EVOO+T (p < .01). Tfam was elevated in Ath and EVOO (P < .01) vs. C, whereas this effect was abolished with EVOO+T. CONCLUSION: Ath enhanced muscle protein acetylation due to over-expression of GCN5 and CD38, whereas these effects were partially corrected with EVOO and EVOO+T. The better preserved NAD+ status could protect against the adverse effects of a high-fat diet on PGC-1α and mitochondrial homeostasis." @default.
