In this study, a PSMA-targeted RNA aptamer labeled with a technetium radionuclide was constructed to explore the potential application of such novel targeting molecules in SPECT/CT prostate cancer imaging. Methods: We selected the PSMA-targeted RNA aptamer A10-3.2 and used succinimidyl 6-hydrazinonic acid hydrochloride (SHNH) as the chelating agent and tricine as the co-ligand to label the aptamer with 99mTc. The in vitro and in vivo biological properties of the labeled aptamer, including the stability, specificity, internalization rate, biodistribution and SPECT/CT imaging, were measured. Results: The uptake of the 99mTc-labeled aptamer A10-3.2 by LNCaP (PSMA+) cells was significantly higher than that by PC3 cells (p < 0.0001), which indicated a good targeting specificity. After intratumoral injection, 99mTc-SHNH-A10-3.2 had a longer tumor retention time and less gastrointestinal and thyroid uptake than technetium sodium sulfate (Na99mTcO4). After tail vein injection, the tumor was partially ingested, and the main excretion routes were through the kidneys and hepatobiliary system. Conclusions: Our data demonstrated that the 99mTc-labeled aptamer A10-3.2 has a good targeting specificity and relatively high stability in vitro, but its in vivo biological properties and tumor uptake need to be further improved." @default.
