Heat shock protein 70 (HSP70) is a cancer biomarker whose level is increased in a plethora of cancerous tumors. Therefore, this work is focused on the development of cost-effective label-free biosensing over a novel plastic chip electrode (PCE) platform for highly sensitive and selective detection of the HSP70 biomarker. The gold-coated surface of PCE was modified with 4-aminothiophenol and glutaraldehyde linking chemistry, and further immobilized with a bioreceptor anti-HSP70. Further, sensing experiments were performed using differential pulse voltammetry. The results confirmed the successful surface modifications and immobilization of the anti-HSP70 antibody on the PCE platform. PCE electrochemical biosensor displayed the highly sensitive detection of the HSP70 in the wide linear range of 0.01-1000 ng/mL and a low limit of detection (LOD) of 3.5 pg/mL at 3σ. Also, HSP70 detection from the serum samples with excellent sensitivity, accuracy and specificity was demonstrated.
