The anticancer property of Marizomib (MARI) encapsulated in poly (lactic-co-glycolic acid) (PLGA) nanoparticles ([email protected]) in vitro by using hepatocellular carcinoma cell lines has been established. Further, the morphological examination, hydrodynamic size and zeta potential of the nanoparticle were examined by the high-resolution transmission electron microscopy (HR-TEM) and dynamic light scattering (DLS) method. The MTT assay has shown a significant inhibitory impact of [email protected] on HCC cancer cells (Caco-2 and HepG2). Furthermore, an investigation of the mechanism showed that apoptosis in cancer cells has performed by anticancer activity, and it was confirmed by various biochemical staining (EdU incorporation assay, AO-EB staining, nuclear staining, and mitochondrial membrane potential assay) methods. This result suggests the decrease HCC treated cells in the mitochondrial membrane potential, which validates the mitochondrial pathway mode apoptosis. Remarkably, [email protected] has confirmed to suggest that the noticeably greater animal safety profile of the MARI is highlighted as prospective candidates for HCC cancer treatment using these [email protected] and deserve further clinical assessment." @default.
