Studi ini bertujuan untuk mengaplikasikan teknik PCR (Polymerase Chain Reaction) untuk mendeteksi daging tikus dalam bakso yang dijual di pasar traditsional. Sampel berupa daging dan bakso tikus (Rattus norvegicus) sebagai control positif, daging sapi sebagai diferensial spesies dan 5 bakso dari berbagai pasar tradisional di Yogyakarta. Pasar tradisional tersebut adalah Bringhardjo, Demangan, Colombo, Condong Catur dan Kranggan. Deoxyribo Nucleotide Acid (DNA) dari sampel diekstrasi dengan kit (Geneaid A‚®, USA) dan amplifikasi DNA menggunakan primer spesifik (5' AAA TCC AAC TTA TAT GTG AAA ATT CAT TGT 3' dan 5' TGG GTC TTA GCT ATC GTC GAT CAT 3') target gen 12S ribosomal RNA. Hasil PCR dianalisis dengan 3,5% gel agarose dan divisualisasi dengan UV transilluminator. Produk PCR yang ditargetkan adalah 96 base pair. Hasil studi menunjukkan PCR dapat menjadi metode untuk mendeteksi pemalsuan daging tikus dalam bakso dengan cepat dan akurat. Namun demikian, sampel bakso tidak mengandung daging tikus. Semua sampel bakso yang dikoleksi dari lima pasar tradisional di Yogyakarta bebas dari pemalsuan daging tikus. Meatball is a very popular meat food products in Indonesia. The adulteration of rat in beef meatball is frequently occurring. This study was aimed to apply PCR technique on the detection of rat in beef meatball from traditional market. In this research, the samples used were rat meat and rat meatball (Rattus norvegicus) which serve as positive control, beef as differential species and five meat-ball from different traditional market in Yogyakarta. These traditional markets are Bringhardjo, Demangan, Colombo, Condong Catur and Kranggan. Deoxyribonucleic acid (DNA) was extracted using kit (Geneaid A‚®, USA) and was then amplified with specific primer with gene target 12S ribosomal RNA (5' AAA TCC AAC TTA TAT GTG AAA ATT CAT TGT 3' and 5' TGG GTC TTA GCT ATC GTC GAT CAT 3'). After that, this PCR products were running by electrophoresis with 3.5% agarose gel. The PCR products were then visualised by UV transilluminator. The targeted PCR product is 96 base pair. The result showed PCR is a fast and accurate method to detect falsification rat meat in meat ball. However, non-of the samples collected contains rat meat. All the meatball sample taken from five traditional markets in Jogjakarta are free from rat meat substitution." @default.
