- W3204351471 abstract "Solid-state discovery and characterization of new active pharmaceutical ingredients is one of the core disciplines during drug development and commercialization in the pharmaceutical industry. The importance of rational solid form selection among polymorphs, solvates, salt forms, and cocrystals is mainly due to their respective relative and thermodynamic stability which ultimately impacts pharmaceutically relevant properties like solubility and bioavailability. Furthermore, different solid forms often feature different particle shapes which in turn affect downstream processes like solid/liquid separation, washing, and drying as well as subsequent drug product formulation steps during large-scale manufacturing. This chapter focuses on solid form monitoring, quantification, and process modeling and is divided in two sections. The first part provides an overview of various process analytical technologies which are used for solid form monitoring during pharmaceutical processing with the corresponding advantages and limitations. The second part is dedicated to advanced quantification approaches and their application to process modeling, including a data-driven model to correlate in situ chord length distribution ( CLD ) measurements with particle size distributions ( PSDs ) data that have been characterized offline." @default.
