• W3209348360 abstract "Parasetamol is a widely-used analgesic because it has antipyretic and analgesic effects and is preferred from children to elderly. Although oral paracetamol is used commonly, in some cases such as patients with unconsciousness, other alternative routes may be required. This study aimed to evaluate the pharmacokinetic properties of supralingual (SL) paracetamol embedded in polyvinyl alcohol (PVA). In the first part of the study, paracetamol loaded nanofiber surface was produced by needle electrospinning method and it was used in the experimental animal study. 12 wt % PVA/distilled water solution was used in the production of nanofiber structure. A total of 24 healthy white New Zealand rabbits divided as controls (5 mg parasetamol, given orally), SL paracetamol (5 mg paracetamol, embedded in PVA nanofibers) and intravenous paracetamol (5 mg paracetamol). The conductivity and viscosity of the solution containing 12% PVA/distilled water was determined as 408 μS/cm and 1,42 Pa s, respectively. With the addition of paracetamol to 12 wt % PVA/distilled water solution, the conductivity of the solution decreased but the viscosity increased. On the other hand, pharmacokinetic parameters werefound similar to oral administration (oral solution) except t max , and bioequivalent to oral solution. SL paracetamol loaded in PVA nanofibrous surface can be a good alternative for the clinical conditions in which patients with unconsciousness or dysphagia." @default.
