- W3217039200 abstract "Pulmonary arterial hypertension (PAH) is a devastating condition affecting the wall and endothelium of pulmonary microvessels, resulting in their partial/total obstruction. Despite current combination of vasodilatory therapies, PAH remains incurable. There is an urgent need for innovative methods to access patient material and unravel progression of PAH. A ground-breaking method to isolate pulmonary arterial endothelial cells (PAECs) from the balloon located at the tip of Swan-Ganz pulmonary artery catheter has been developed and moved forward. We aim to report our incremental experience in optimizing the initial methodology and phenotyping PAECs along in vitro subcultures. PAECs, harvested from Swan-Ganz catheters during routine diagnostic or follow-up right heart catheterization of patients with pulmonary hypertension, were phenotyped by immunofluorescence and flow cytometry using antibodies against specific endothelial markers. From the 132 Swan-Ganz catheters collected, we observed growing cells from 56 procedures, expanded and stored cells from 23 procedures. The endothelial phenotype remained stable in early subcultures, whereas cell hypertrophy, occurrence of myofibroblast-like cells, mitigated expression of specific endothelial markers (CD31, VE-cadherin, von Willebrand factor) and reduced scavenger receptor-mediated acetylated-LDL uptake were observed in advanced subcultures. Our results highlight the unique opportunity to obtain homogenous and stable subcultures of PAECs at diagnosis and follow-up. Within the era of precision medicine, it opens promising perspectives for patients suffering from rare pulmonary vascular diseases." @default.
