• W3217593380 abstract "<b>Aims:</b> IL-17 is involved in the recruitment of eosinophils and neutrophils. Although its inhibition is capable of modulating inflammatory and functional responses in asthma and COPD. <b>Objectives:</b> Evaluate the effects of anti-IL-17 in pulmonary mechanics and inflammatory pathways in an experimental ACO model. <b>Methods:</b> BALB/c mice were sensitized with ovalbumin (OVA) or elastase (ELA) followed by treatment with or without anti-IL-17. We evaluated respiratory mechanics and bronchoalveolar lavage fluid (BALF). <b>Results:</b> The treatment with anti-IL-17 in OVA anti-IL-17 (Rrs 8.4 ± 1.4 cmH2O/L/s) (Ers 4.7±0.9 ml/cmH2O), ELA anti-IL-17 (Rrs 4.9±1.0 cmH2O/L/s) (Ers 3.6±1.4 ml/cmH2O) and ACO anti-IL-17 (Rrs 11.1±3.4 cmH2O/L/s) (Ers 4.3±1.0 ml/cmH2O) groups reduced the resistance and the elastance of the respiratory system when compared to OVA (Rrs 34.2 ± 10.5 cmH2O/L/s) (Ers 12.2±3.1 ml/cmH2O), ELA (Rrs 12±1.6 cmH2O/L/s) (Ers 8.7 ± 1.6 ml/cmH2O) and ACO (Rrs 37.7±8.3 cmH2O/L/s) (Ers 9.4±0.8 ml/cmH2O) groups, p<0.05. There was also reduction in the number of total (3.5±1.0) and differential inflammatory cell count neutrophils (0.2±0.0), eosinophils (0.9±0.4), macrophages-104cells/mL in BALF (3.6±0.8) in the ACO anti-IL-17 group when compared to ACO, p<0.05 for all comparisons. There was a reduction in the number of exhaled NO in the OVA anti-IL-17 (9.7±1.1), ELA anti-IL-17 (8.7±1.2) and ACO anti-IL-17 (9.4±2.5) treatment groups when compared to control groups OVA (24.1±4.3), ELA (9.6±2.1) and ACO (13.7±4.4), p<0.05 for all comparisons. <b>Conclusions:</b> Inhibition of IL-17 contributes to control of Th2/Th17 inflammation and oxidative stress in an ASTHMA-COPD overlap syndrome (ACO) model." @default.
